Paul Theroux nasceu em Medford, no Massachusetts, em 1941. O pai era franco-canadiano e a mãe italiana, e Paul era um dos sete irmãos. Frequentou as Universidades do Maine e, posteriormente, do Massachusetts. O curso de Escrita Criativa que realizou com o poeta Joseph Langland fê-lo descobrir que escrever era o que queria fazer na vida. Viveu em Itália, onde foi leitor; no Malawi, onde também ensinou e esteve envolvido no golpe de Estado que tentou depor o então presidente-ditador; em Singapura e no Uganda, onde deu aulas de Inglês e não só conheceu a sua futura mulher como também encontrou, pela primeira vez, V.S. Naipaul (que viria a ser seu grande amigo e mentor). Paul Theroux vive atualmente entre Cape Cod e o Havai.
A par das colaborações regulares que manteve ao longo dos anos com as revistas Playboy, Esquire e Atlantic Monthly, escreveu dezenas de romances (alguns adaptados ao cinema), ensaios e alguns dos melhores livros de viagens de sempre, como O Velho Expresso da Patagónia, Comboio Fantasma para o Oriente e O Grande Bazar Ferroviário, todos publicados pela Quetzal.
